P-46: Proliferative Activity of Follicular Cells in Adult Mouse Ovary Following Administration of Exogenous Nicotine

Background: Nicotine can induce sub fertility and infertility in females. The aim of this study was to determine the effect of nicotine on follicular cells proliferative activity in mouse ovary. Materials and Methods: Female adult NMRI mice were divided into two groups. Animals in control group received vehicle, however in experimental group, animals were treated with 0.4mg/kg of nicotine for 14 consecutive days, intraperitoneally. All animals were dissected on 15th day and all ovaries were dissected and evaluations were made by Ki-67 IHC Staining for studying of proliferative activity of the cells in follicles. Statistical analysis was performed using t test. Results: The mean numbers of Ki-67 positive cells in follicles of control group were 9.14 ± 7.06 in primary follicles and 33.80 ± 18.35 in secondary follicles and no Graafian follicle was seen. There was not significant statistical differences in numbers of Ki-67 positive cells of nicotine treated group in compare with control group. Conclusion: The results from this study suggest that administration of 0.4 mg/kg nicotine for 14 consecutive days has not effect on proliferative activity of the cells in adult mice ovary
